Introduced in 2007, Service 5 is an inter-town route connecting the East and Bukit Merah through the Central areas of Orchard and Novena. The route serves residential estates around Pasir Ris, Flora Drive, Upper Changi, Simei, Bedok Reservoir, Tiong Bahru and Bukit Merah with connections to the City areas of Havelock, Paterson and Novena. The route plies an express sector along the Pan-Island Expressway (PIE) between Bedok Reservoir and Novena district, offering improved express connections between these areas.

Serving many inter-town connections, the route is frequently crowded during peak hours. Its express sector is especially well patronized as a direct connection between Bedok Reservoir, Jalan Toa Payoh and the City areas.

• 2011 (27 Jun): Designated wheelchair-accessible bus service (WAB)
• 2011 (28 Aug): Amended to ply Jln Tenaga and Jln Damai with the partial closure of Kaki Bt Ave 1 to facilitate the construction of Kaki Bukit MRT station on the Downtown MRT Line
• 2016 (24 Apr): Amended to ply the reinstated Kaki Bukit Ave 1, skipping Jln Tenaga and Jln Damai
• 2018 (18 Nov): Start of operations under Bukit Merah Bus Package (Tendered Contract)
