Service 54 connects the housing estates of Bishan and Marymount with the Central areas of Novena, Scotts Road, River Valley and Chinatown. Similar to Service 56, it serves Bishan Street 13 in both directions. From the stretch of River Valley Road it serves, it is the only service which links to Orchard, Newton and Novena.
Service hours are extended on Saturdays to benefit late-night commuters returning home from Chinatown and Clarke Quay.

History:
- 1993 (05 Dec): Introduced between Bishan Int and New Bridge Rd Ter with the integration of Services 104 and 134 under TransitLink Network Integration Exercise Phase 14
- 1994: Last bus from New Bridge Rd Ter revised to 2330 hrs daily
- 1997: First bus from Bishan Int brought forward to 0545 hrs on weekdays
- 1997 (07 Dec): Amended to ply Upp Pickering St, South Bridge Rd and Kreta Ayer Rd with the partial closure of New Bridge Rd to facilitate the construction of Chinatown MRT station on the North East MRT Line
- 2000 (20 Feb): Serves additional bus stop opposite Blk 333 along Kreta Ayer Rd
- 2000: First bus from New Bridge Rd Ter extended to 2400 hrs on Saturdays
- 2002 (20 Apr): Amended to ply the reinstated New Bridge Rd, skipping Upp Pickering St, South Bridge Rd and Kreta Ayer Rd
- 2006 (05 Nov): Amended to ply Bishan St 13 and Bishan St 11
- 2009 (03 Dec): Designated wheelchair-accessible bus service (WAB)
- 2018 (10 Mar): Exended to Kampong Bahru Ter with the closure of New Bridge Rd Ter. First bus from Kg Bahru Ter brought forward to 0610 hrs daily
- 2020 (08 Nov): Skips the bus stop opposite SLF Cplx along Thomson Rd to facilitate the construction of the North-South Corridor (NSC)
- 2023 (08 Jan): Temporarily skips the bus stop outside Newton Stn Exit B along Scotts Rd due to upgrading works
- 2023 (27 Aug): Resumes serving the bus stop outside Newton Stn Exit B along Scotts Rd after completion of upgrading works
