City Direct Service 646 offers residents of Tampines North and Tampines West improved connectivity to the Central Business District areas of Marina Bay and Shenton Way. The route plies express via Simei Ave, Xilin Ave, and East Coast Parkway (ECP), providing a direct travel alternative to commuters who typically rely on the rail network and feeder buses.

As a City Direct service, it operates only on weekdays (except Public Holidays), with unidirectional City-bound trips in the mornings, and return trips towards the heartlands in the evenings.

The route was launched on 15 June 2026, as a renumbering of Express Bus Service 513 (Tampines Ave 5 ⇄ Shenton Way), which was discontinued on the same day. The new route extends Express 513 from Tampines Ave 5 to serve Tampines North, plying Tampines Ave 6, Tampines Nth Dr 1, and Tampines St 64, while skipping a pair of bus stops along Tampines Ave 5 originally served by Express 513 (Bus Stop 75121 / 75129, outside Darul Ghufran Mosque).

If the eve of Christmas, New Year’s Day, and Chinese New Year fall on a weekday, evening peak departures will be brought forward to the afternoon. Revised departure times will be provided by the operator closer to the affected date.
