City Direct Service 650 offers residents of Yishun East improved connectivity to the Dhoby Ghaut, Bras Basah, and Shenton Way. The route plies express via Lentor Ave, Seletar Expressway (SLE) and Central Expressway (CTE), providing a direct travel alternative to commuters who typically rely on the rail network and feeder buses.

As a City Direct service, it operates only on weekdays (except Public Holidays), with unidirectional City-bound trips in the mornings, and return trips towards the heartlands in the evenings.

The route was launched on 15 June 2026, as a renumbering of Express Bus Service 850E, with one additional morning trip added, for a total of 9 morning trips at launch. The route remains unchanged. This renumbering exercise also saw other peak-hour express bus services to the City reclassified as City Direct services.

If the eve of Christmas, New Year’s Day, and Chinese New Year fall on a weekday, evening peak departures will be brought forward to the afternoon. Revised departure times will be provided by the operator closer to the affected date.
